| I. Background | |

LGR5 基因（富含亮氨酸重復(fù)序列的 G 蛋白偶聯(lián)受體 5）是 GPCR 家族成員，位于染 色體 12q22-23，編碼 907 個(gè)氨基酸的跨膜蛋白。其胞外區(qū)含 17 個(gè)亮氨酸重復(fù)序列，通過(guò)結(jié) 合 R-spondin 增強(qiáng) Wnt/β-catenin 信號(hào)通路，調(diào)控干細(xì)胞增殖、分化及腫瘤發(fā)生。
LGR5 作為 Wnt 受體的協(xié)同因子，通過(guò)與 Frizzled 和 LRP5/6 受體結(jié)合，促進(jìn)[Wnt/β- catenin 信號(hào)通路激活。在正常組織中，LGR5 表達(dá)局限于腸道隱窩、毛囊等干細(xì)胞區(qū)，參 與組織再生修復(fù)；在腫瘤中，其高表達(dá)與結(jié)直腸癌、肝癌、乳腺癌等密切相關(guān)，作為癌癥 干細(xì)胞標(biāo)志物，促進(jìn)腫瘤侵襲、轉(zhuǎn)移及化療耐藥。機(jī)制上，LGR5 通過(guò)激活 Wnt 通路介導(dǎo) 上皮-間質(zhì)轉(zhuǎn)化（EMT），并抑制 p53 穩(wěn)定性增強(qiáng)耐藥性。 |
