TP53 基因編碼 p53 蛋白,是調(diào)控細(xì)胞周期、DNA 修復(fù)及凋亡的核心腫瘤抑制因子。 R175H 突變是 TP53 第 175 位氨基酸由精氨酸(R)突變?yōu)榻M氨酸(H)的錯義突變,位于其 DNA 結(jié)合域的關(guān)鍵功能區(qū)。該突變導(dǎo)致 p53 蛋白構(gòu)象改變,喪失與 DNA 結(jié)合的能力, 無法正常啟動細(xì)胞周期阻滯或凋亡程序,從而促進(jìn)腫瘤細(xì)胞異常增殖、轉(zhuǎn)移及化療耐藥。 |
